The plot of the movie this time does not entirely focus on racing but on an international espionage as well. Mater (Larry the Cable Guy) is approached by a British secret agent who is also known as the world's greatest spy, Finn McMissile (Caine), believes that there is a conspiracy cooking behind the World Grand Prix race which he has learned from a fellow spy, Rod "Torque" Redline. Initially, McMissile thought of Mater as fellow secret Agent living in the America that is why he asks his assistance. He then also seeks McQueen's help alongside his aid agent, Holley Shiftwell (Mortimer) and Mater on a dangerous mission to crack the case of the World Grand Prix conspiracy.
Mater is divided between helping his best friend win a race of a lifetime and helping the secret spies stop the villain Professor Z, short for Zundaap, the evil mind behind the attempt to sabotage the World Grand Prix. The mad but brilliant professor is in the top list of the Most Wanted by the officials as he creates dangerous weapons he uses for evil.
